Tn6375 is an 18.8-kb transposon, found on plasmid p447-IMP in Klebsiella pneumoniae strain 447. It was derived from the Tn3-family unit transposon Tn1722 with an IRL–tnpAR–res–mcp (methyl-accepting chemotaxis protein)–IRR structure and it had undergone two major evolutionary events: (i) the insertion of In73 at a site within res, resulting in its truncation from 120 bp to 99 bp; and (ii) the integration of ISEcp1:IS1R–blaCTX-M-14–IS903D (at a site within mcp, splitting it into two separate parts), in which an intact IS1R element was inserted into ISEcp1 (interrupting tnpAISEcp1 into two separate parts).

Tn6378 was derived from Tn1696 after four major evolutionary events: (i) interruption of IRR into two parts by IS5075; (ii) loss of mer; (iii) insertion of In73 [IRi–intI1–blaIMP-8–aacA4′-3–tniABQR–IRt] at the same position within res, truncating it into a single 83 bp remnant; and (iv) insertion of the macAB–tolC region (ABC-type macrolide-resistance efflux system) downstream of In73.
